About the Role

Antibody-drug conjugates (ADCs) show great promise in the clinic but there are several unanswered questions that limit our understanding of their full biological activity. These include on-target biology questions, such as how to select the best cellular model(s) and pharmacodynamic readouts to assess activity, and how tumor and non-tumor cells interact in the complex tumor environment to modulate ADC function. Other questions are related to molecular properties and how various parts of the molecule (antibody, linker, payload etc.) contribute to desired and undesired activity. Finally, there are questions around safety risks, such as how to efficiently study on-target, off-tumor risks, and how to use in vivo safety findings from clinical ADCs to inform cellular model selection and therapeutic index in vitro. This postdoctoral proposal will leverage unbiased profiling through high-content imaging (HCI) and transcriptomics to build signatures for a toolbox of clinically validated ADCs and address some of these questions.

Responsibilities:

  • Designing, planning, and conducting hypothesis driven translational research to define a novel ADC cellular assay toolbox.

  • Increase mechanistic understanding of factors affecting bystander effect of intact ADCs and payloads through well-designed in vitro experiments and integration of the most recent literature.

  • Evaluating, and implementing novel translatable cell models improving the correlation between cytotoxicity of ADCs measured in vitro and efficacy measured in preclinical models and patients.

  • Providing interpretation of data and communicating/reporting results and recommendations to team members, key stakeholders, and collaborators.

  • Using modelling approaches investigate whether in vitro-in vivo correlations can be established.

  • Disseminate the results at internal and external meetings, and through publications in high impact journals.

Qualifications

  • PhD in Pharmaceutical Sciences, Biology, Biochemistry, Pharmacology, Biomedical Sciences or equivalent. Degree must be received in the last three years, or by Jan 2025 for current students.

  • Theoretical knowledge and practical experience in cellular assay development, ideally using biologics and/or ADCs.

  • Hands-on experience in culturing cell lines, spheroids and/or organotypic models.

  • Experience with immunostaining, microscopic, and transcriptomics techniques.

  • Ability to independently determine study design, execution, data analysis and interpretation of in vitro data. A strong interest in practical in vitro studies and a keen interest in troubleshooting will be vital to ensure successful completion of the project.

  • Strong scientific interest with confirmed capability to perform independent research indicated by a proven scientific track record.

  • Display a high degree of scientific curiosity and creativity required to tackle complex investigational hypotheses. A keen interest to acquire in-depth knowledge of literature is crucial for a successful project.

This is a two-year duration-based role that has eligibility for severance. The anticipated base pay for this role is $95,000 per year.

The Company maintains highly competitive, performance-based compensation programs. Under current guidelines, this position is eligible for an annual performance bonus in accordance with the terms of the applicable plan. The annual performance bonus is a cash bonus intended to provide an incentive to achieve annual targeted results by rewarding for individual and the corporations performance over a calendar/performance year. Bonuses are awarded at the Companys discretion on an individual basis.
