RESEARCH DESCRIPTION: The Rsch Pro 6-Biomedical Engineering will perform quantitative experiments related to cell-free transcription-translation, cell-free phage synthesis and phage genome engineering, biophysics measurements using fluorescence microscopy and a quartz crystal microbalance with dissipation (QCMD) with cell-free reactions. The Rsch Pro 6-Biomedical Engineering must know how to prepare and quantitively use cell-free transcription-translation systems from E. coli, use a QCMD, perform quantitative measurements using fluorescence microscopy, SDS PAGES, DNA electrophoresis, DNA technologies (DNA assembly, sequencing). JOB DESCRIPTION: 80% Research This position will develop research plans, and solve highly complex problems through the original design of protocols, policies, and procedures. The research subject is proposed by the incumbent and decided in agreement with supervisor to be inline with general orientation with other research activities within the Noireaux biomedical physics research group. Prepare and quantitively use cell-free transcription-translation systems from E. coli Use a quartz crystal microbalance with dissipation Perform quantitative measurements using fluorescence microscopy, SDS PAGES, DNA electrophoresis, DNA technologies (DNA assembly, sequencing). Analyze, interpret, plot the data for all the experiments 10% Communicating Research Results Maintaining familiarity with cutting-edge results and methods. Attending meetings, seminars and conferences Attending weekly and monthly lab meetings Train younger lab members when needed Communicating research to the broader community Writing papers and submitting them for publication in scientific journals Presenting results at conferences (both domestic and international) 10% Lab maintenance Ordering research supplies Lab cleaning Equipment and Instrument maintenance Other duties of a similar scope as assigned. Qualifications REQUIRED QUALIFICATIONS: Ph.D. in biophysics, biochemistry, synthetic biology or related field. The Rsch Pro 6: - must have shown expertise to prepare cell-free transcription-translation systems from diverse E. coli strains with capabilities of at least 3 mg/ml of protein synthesis. - must have shown capabilities to utilize acoustic liquid dispensers, plate readers, spectrometers, to perform cell-free gene expression reactions and quantitative fluorescent reporter protein measurements. - must have shown abilities to make SDS PAGES for protein analysis, DNA assembly for cloning (Gibson, MoClo) and sequencing. - must know how to use a cell-free gene expression on quartz crystal microbalance with dissipation for membrane biophysics measurements. - must know all the fundamental phage assays: plaque assay, spotting assay, phage infection assay. - must know how to use fluorescence microscope and make quantitative fluorescence measurements of cell-free gene expression reaction samples.
